研究概要	1.大脳皮質は原則的に6層から構成され、錐体細胞はその主要な構成ニューロンであり、錐体細胞は皮質外へ出力するばかりでなく、皮質局所に軸索側枝を多数出力し皮質内の情報処理にも重要な機能を持つと考えられる。大脳皮質出力ニューロンを逆行性に細胞体・樹状突起をGolgi染色様に標識する方法により前もって出力ニューロンを標識したラットの大脳皮質スライス標本を用いて、微小ガラス電極を各層のニューロンに刺入し、基本的な電気的性質を記録し、分類した。記録後、ニューロンにBiocytinを注入し、スライス標本を固定する。組織学的な検索として、まず三重蛍光法を用いてBiocytinを注入したニューロンが興奮性あるいは抑制性であるか検討した。Biocytinを注入したニューロンと特にその出力である神経軸索をAvidin-biotinylated peroxidase complex法によって青黒色に染色した。この神経軸索の入力先として逆行性に標識された出力ニューロンをPeroxidase anti-peroxidase酵素免疫染色法で赤く染色した。III層錐体細胞の出力はV層錐体細胞へ多く入力してたが、VI層錐体細胞へはその4分の1しか入力していなかったことを報告した。さらにVI層の錐体細胞への入力を調べII/III層およびV層の錐体細胞からの入力は少ないが、IV層・VI層からの入力が多いことを見いだした。 2.大脳皮質の投射ニューロンをその軸索末端からアデノウィルスベクターに感染させ、膜移行シグナルを導入したGreen fluorescent protein (GFP)を発現させ、Golgi染色様に逆行性標識することに成功した。 3.大脳皮質GABA作動性の内在性抑制ニューロンをGolgi染色様に染色する手法を、トランスジェニックマウスを利用して作ろうとしている。現在、GABAニューロンのマーカーであるParvalbuminとCalretininというカルシウム結合タンパク質のプロモーター部位をクローニングし、特異性の検討をしている。これらのGABAニューロンに2と同じく膜移行シグナルを導入したGFPを発現させる予定である。
