研究概要

本年度は主に、1)海馬各部位のニューロン構成のstereologyによる定量解析及び2)マウス歯状回hilusのmossy cellの形態学的、免疫細胞学的解析、更に、電気生理的解析を進めた。
1)蛍光抗体法で染色した40-50μm厚のスライスを共焦点レーザー走査顕微鏡(現有)で観察検討し、少なくとも細胞体レベルで免疫染色浸透性が均一な条件を確立した。そのような染色条件を満たす切片標本を用いていわゆる非錘体細胞をいくつかの免疫細胞学的に同定できる化学的マーカーに基づいて分類しそれらをdisector法で解析し、各部位のさらに、層による細胞構成の差異を明らかにした。
2)カルシウム結合蛋白であるcalretininの海馬形成での分布を歯状回においてラットとマウスで比較し、著しい種差を明らかにしたが、その最も大きな差異を示したhilar mossy cellの解析を更に進め、以下のことを明らかとした。ラットではcalretinin陰性のhilar mossy cellがマウスではcalretinin陽性であり、このようなcalretinin陽性mossy cellが背側歯状回門には見られず、腹側歯状回門に集中している。また、背側歯状回門には、腹側歯状回門よりかなり少数ではあるが、mossy cellと同定できるcalretinin陰性細胞が存在している。すなわち、マウスではcalretinin含有の点で、mossy cellは多様であり、背側部と腹側部で異なっていた。さらに、構造的にも背側部と腹側部ではthorny excresencesの複雑さで異なっていた。また、スライスを用いた電気生理的解析により、マウスhilar mossy cellはラットhilar mossy cellと異なった電気生理的性質を持っていることを明らかにした。
