ドーパミン受容体ノックアウトマウスによる脳機能の研究

研究概要	脳機能解明の最終目標は、ヒトの脳機能である。しかし、ヒトは実験対象とはならないから、その研究戦略には工夫を必要とする。一方、アゴニストやアンタゴニストの受容体に対する作用の研究がヒト個体レベルで豊富に存在するドーパミン神経系の研究は非常に興味深い。本研究の目的は、ES細胞を用いた2段階相同組換えにより、マウスのドーパミン受容体遺伝子をヒトの遺伝子で置換することにより、ドーパミン受容体をヒト型にし、ヒトに作用し、マウスに作用しない、アゴニストやアンタゴニストの作用を検討する。これまで薬理学的、生理学的に解明されてきたドーパミン神経系の機能を、ドーパミン受容体に注目し、遺伝子を欠損させた突然変異ばかりでなく、ヒト型マウスを作成し、高次の脳機能を、一挙に分子レベルのメカニズムから説明しようとするものである。本年度は5種のドーパミン受容体遺伝子のうち、D1R,D2R,D4Rの遺伝子を単離し、D3R,D5Rについては現在進行中である。また、D4Rについては、第一段階のキメラマウスが得られた。D4受容体と高い親和性を持つ非定形型抗神経病薬であるクロザピンに対するヒトとマウスとの親和性の違いが示唆されている。また遺伝子多型と分裂病との関係を知るため、多様な繰り返し構造を持つヒトD4受容体遺伝子をマウスD4受容体遺伝子座に導入する実験を行う。
