研究概要

本研究の目的は、組み換えアデノウイルスを用いることにより、多くの高等動物で汎用できるウイルス感染における自然宿主での細胞性免疫機構の解析法を開発することである。各種動物において以下の様な知見を得た。
(1)昨年度作出に成功したイヌジステンパーウイルス(CDV)膜蛋白(H)遺伝子の組み換えアデノウイルスを用い、以下の成果を得た。CDV-H組み換えアデノウイルスを用いて細胞障害試験を樹立するため、標的となる自己細胞を確立するため、皮膚からの細胞株樹立を試みた。その結果複数のイヌからの樹立に成功し、標的細胞の作製法はほぼ確立した。さらに、その他の条件を決定し、細胞障害試験の確立を試みている。
(2)細胞障害活性及び液性免疫能を誘導するエピトープが牛疫ウイルスのN蛋白に存在することが明らかとなった。H蛋白の免疫により長期に免疫賦与される事も明らかになった。また、immunestimulating complex(ISCOM)の利用により、組み換えH蛋白でも細胞性免疫賦与が可能であることが明らかとなった。
(3)牛白血病ウイルス(BLV)の主要組織適合性遺伝子複合体(BoLA)クラスII分子には白血病に抵抗性を示すBoLA-DR3アリルが存在することが明らかとなった。羊を用いた接種実験で、これを確認したため、現在BLVの組み換えアデノウイルスを作製して、さらに詳細な解析を試みている。
