疎水化多糖類被覆組み替え癌遺伝子タンパクによる抗腫瘍性ワクチンの開発

研究概要

組み替えHER2蛋白とコレステロール疎水化多糖類プルランの複合体(CHP-HER2)によりHLA-A2402陽性及び陰性の健常人より調整された末梢血CD14^+細胞由来樹状細胞を前処置した。処置18時間後に、これらの樹状細胞のHLA-A2402拘束性HER2p63ペプチド特異的CD8^+CTLクローンに対する感受性を検討した。CHP-HER2前処置のHLA-A2402陽性樹状細胞のみがCTLクローンにより殺傷され、コントロール複合体で処理された樹状細胞及びHLA-A2402陰性樹状細胞は全く感受性を示さなかった。さらにHLA-A2402陽性樹状細胞をCHP-HER2複合体、CHP、HER2蛋白のみ等にて前処置した後にHER2p63特異的CTLクローンに対する刺激能を検討した。CHP-HER2によって前処置された樹状細胞のみがCTLクローンを刺激しインターフェロンγの産生を促したが、CHP、HER2蛋白、CHPとコントロール蛋白の複合体により前処置された樹状細胞は何ら刺激活性を示さなかった。CHP-HER2前処置樹状細胞のHER2p63ペプチド提示に関わる分子を明らかにすべく、CHP-HER2前処置樹状細胞を各種inhibitorで処理し、ペプチド提示能に対する影響を検討した。サイトカラシン D、ブレフェルディン A、サイクロヘクサマイド、及びラクトシスティン前処置樹状細胞はペプチド提示能を消失した。また、CHP-HER2前処置HLA-A2402陽性樹状細胞により、自家末梢血リンパ球を感作し、HER2p63特異的HLA-A2402拘束性CTLクロンーンを樹立できた。
