研究概要

1. Invasinに関する基礎的研究
昨年までに同定したinvasin蛋白の細胞内侵入のために必要最低限部分蛋白をGST-fusion蛋白として作成・精製した。これをポリマーもしくはモノマーの形でヒト線維芽細胞や悪性グリオーマ細胞の培養液中に加えて、細胞内への侵入の状態を観察した。この時に、integrin bの会合が必須であることが明らかとなった。また、この系に細胞骨格の重合を変化させる各種薬物を加えてその影響を観察した1ところ、アクチンの重合と微小管の集合がinvasinによって誘導される細胞貪食作用の制御にかかわることが明らかとなった。
2. Invasin付加可能なマイクロスフィアーの開発
難水溶性抗癌剤の封入したマイクロスフィアを開発する基礎実験として、生体内分解高分子であるポリ乳酸を用いて難水溶性テオフィリンを含むマイクロスフィアの作成に成功した。封入効率も上昇し、徐放性も得られた。この手法を応用してポリデプシペプチドを用いて抗癌剤含有マイクロスフィアーを作成する方法もほぼ完成した。
このマイクロスフィアーとグルタチオンを反応させてGST蛋白と結合可能なスフィアーを作成する条件を決定しつつある。
