コラーゲン結合接着因子(CMP,RGD-CAP)の活性部位を用いた生体材料の開発

研究概要

コラーゲン結合蛋白(CAP)は組織発生、増殖、分化、修復に重要な役割を果たしている。私たちは軟骨コラーゲン線維画分より、RGD-CAPおよびCMP(Matrilin1)などのCAPを精製した。本年度は、RGD-CAPとCMPを各種の細胞培養系に添加して、接着および伸展に及ぼす影響を検討した。そしてこれらが、ウサギ関節軟骨細胞、ウサギ線維芽細胞、ニワトリ軟骨細胞、ヒト軟骨細胞およびヒト線維芽細胞の接着と伸展を促進することを明らかにした。しかしヒト歯髄細胞の接着には影響しなかった。またRGD-CAPの細胞接着活性部位としてC-末端に存在するRGD配列はその活性に必須ではないことが明らかとなった。さらにRGD-CAPとCMPはコラーゲンに対するインテグリンであるα1β1インテグリンと結合することが明らかとなった。組み換え体RGD-CAPを作成して各種の断片の接着促進活性を比較して、RGD-CAPの接着にはその4つの分子内繰り返し構造のうち2個以上の存在が必須であることが明らかになった(大野ら、日本骨代謝学会報告1998)。またCMPはα1β1インテグリンと直接結合し、その接着促進活性にはコラーゲンの存在が必要であることを明らかにした。CMPは軟骨にしか発現していないので、CMPは軟骨分化に特別な役割を果たしていることが示唆された。なおCMPについての私たちの成果はJ.Biol.Chem.(May,印刷中)に報告した。
