コラーゲン結合接着因子(CMP,RGD-CAP)の活性部位を用いた生体材料の開発

研究概要

本研究において、Matrilin1(CMP)が2型コラーゲン存在下でalpha1beta1integrinと結合することが判明した。(J.BIOL.CHEM.1999)。またRGD-CAPの接着促進活性にもalpha1beta1integrinが必須であることを証明した。(B.B.A.1999)。
さらに私たちは、コラーゲン結合接着因子であるRGD-CAPの活性部位を決定するために、アミノ末端から蛋白を削除した断片およびC-末端から蛋白を削除した断片を数種類作成した。その結果、1)C-末端近傍に存在するRGD配列を持たない断片でも接着促進活性を80%以上保持していること、2)分子内に4つ存在するうちの2つ以上の繰り返し配列の存在が接着促進活性に必須であること、3)その繰り返し配列は4つのうちのどれでも2つ以上あれば活性があることを証明した。
一方、Matrilin1(CMP)とMatrilin3の活性部位を決定するためにウサギ軟骨細胞培養系から両者のcDNA断片をクローニングした。その結果、ウサギMatrilin3のEGF様配列の数が、これまで報告されているマウスのもの(4つの繰り返し)とは異なること(5つある)が明らかになった。
