アポトーシスとIL-1産生抑制を引き起こすアクチビンを用いた歯周炎予防法の開発

研究概要	これまでに、数種類のアクチビン受容体がクローニングされ、cDNA配列の結果からどれもがセリン/スレオニン型の受容体であることが明らかとなった。このようなタイプの受容体はこれまでにない未知の細胞内情報伝達機構に関与していることが予想されている。我々は、TGF-βスーパーファミリーに属するアクチビンAがマウスB細胞ハイブリドーマのG1期での細胞周期の停止とアポトーシスを誘導することを見い出し、その発現機構を解析してきた。本研究では、アクチビンがアポトーシスを発現する際に関与している受容体の解析とアクチビンが受容体に結合した後の情報伝達系を調べることを目的としている。そこで、本年度は、アクチビンによる細胞周期の停止とアポトーシス発現の情報伝達系におけるアクチビンI型受容体の役割を検討した。アクチビンに対して感受性の高いB細胞ハイブリドーマHS-72細胞にアクチビンI型受容体であるIA(ActRIA)およびIB(ActRIB)の発現ベクターを導入してそれぞれの安定発現株を作製し、それらのG1期での細胞周期の停止とアポトーシス発現について解析した。その結果、ActRIBの発現細胞株(HARIB)ではアクチビンに対する感受性が亢進した。一方、ActRIAの発現細胞株(HARIA)では、アクチビンに対する感受性は低下していた。このことから、HS-72細胞におけるアクチビンの増殖抑制とアポトーシスの情報はActRIBを介して伝達されることが示唆された。さらに、ActRIAはアクチビンのActRIBを介した情報伝達をドミナントに抑制する調節因子として機能していることを示唆する結果が得られた。今後、アクチビンが受容体に結合した後に、アクチビンI型受容体から細胞内への情報がどのように伝達されているかについてより詳細に検討を加えていくつもりである。
