研究概要	従来、抗体の調製には長時間を要し、しかも親和定数で10^<12>・M^<-1>以上の親和力を持つ抗ハプテン抗体を得ることはきわめて困難な課題とされている。そればかりか、目的とする高い特異性を持つ抗体を得るには、適切な感性抗原のデザインと、これを合成する高度な有機化学的手法が要求される。本研究の目的は、こうした問題を一挙に解決すべく、遺伝子レベルで抗体分子の相補性決定領域(CDR)のアミノ酸配列をランダムに変換し、生体のレパートリーには見られない一次構造を有する「人工抗体(超抗体)」のライブラリーを構築し、これによりハプテン抗原に対してきわめて高い特異性と親和力を持つ超抗体を作製しようとするものである。本年度は、本計画の初年度とし、抗デオキシコルチゾール(DOC)抗体(CET-M8)を、DOCの4位にブリッジを導入したハプテンを用い、常法に従い調製した。次いで、これをwild-type抗体とし、γ_1、κ鎖に特異的なプライマーを用いてcDNAを合成した。引き続き、VH-DNA、VL-DNAをリンカー配列DNAと連結してscFvフラグメント(scFv)のDNAを調製し、ファージミドベクターをクローニング後大腸菌にtransformationし、十分な力価を持つファージを得た。本ファージ抗体のDOCに対する反応性をELISA法により検討したところ、4種のクローンに強いシグナルが見られ、目的のscFvがファージに提示されたものと期待される。これらの結果は、次年度以後の研究を進めるのに十分な成果といえる。
