キチン結合性を有する抗真菌蛋白質の探索と構造解析及び抗真菌剤開発のための基礎研究

研究概要

日本産カブトガニTachypleus tridentatusの顆粒細胞から3種の抗菌性ペプチド、タキスタチンA、B、Cを精製した。これらのタキスタチンはグラム陰性菌、陽性菌、真菌に対し広い抗菌活性があり、中でもタキスタチンCは、何れの菌に対しても最も強い活性を示した。また、顕微鏡観察の結果、タキスタチンCは、出芽酵母Pichia pastorisの溶菌を引き起こすことも判明した。タキスタチンCを蛍光ラベルして、P.pastorisに作用させると、細胞壁の、特にキチンの豊富な部分が強く染色され、タキスタチンCは、細胞壁の主にキチンを認識していることが示唆された。さらに、細胞傷害性を調べるため、溶血活性を測定したところ、タキスタチンCは、カブトガニの抗菌性ペプチドの中で最も強い溶血活性を示した。
タキスタチンCは、3個のジスルフィド結合を含む41アミノ酸残基からなり、6個のCysの位置は3種のタキスタチンの間で保存されていた。タキスタチンAとBの配列類似性は40%であったが、タキスタチンCとAおよびBとの有意な相同性は見られなかった。一方、タキスタチンCは、殺虫作用のあるクモの神系毒、μ-agatoxinとcurtatoxinsに相同性が見られた。タキスタチンCは、C末端に特徴的な、両親媒性のβシート構造をとっていることが推定され、これが、抗菌活性に重用な役割を果たしていると思われる。
