制限修飾遺伝子の自己維持機能を利用したプラスミド安定化

研究概要	制限酵素の遺伝子は、同じ認識配列をメチル化し制限切断から保護する修飾酵素の遺伝子に隣接する。「制限修飾遺伝子単位は、外敵DNAを切断することによって細胞を守るための道具である」というのが定説であった。私たちは、「制限修飾遺伝子を持つプラスミドは細胞から失われにくい」という現象を発見し、「細胞から制限修飾遺伝子が失われると、修飾酵素が減少し、新しく作られる染色体上の認識配列をメチル化しきれなくなる。そこを残った制限酵素が切断すると、細胞は殺される。」という「分離後の宿主殺し」モデルを提案した。これらから、「制限修飾酵素遺伝子が、トランスポゾンやウイルスのような独自の遺伝単位である」という「利己的な遺伝子説」「ゲノム病原体説」を提唱した。制限修飾遺伝子のこの「利己的な」自己維持作用を解明し、それに連結することによってプラスミドの安定な維持を図ろう、という本研究の目的は、次のように達成された。安定化効果は、大腸菌でEcoRI,EcoRV,PaeR7,PvuIIの場合に観察された。 (1)プレート法によって、細胞死・安定化に影響する大腸菌の遺伝子を多数探しあてた。 (2)制限修飾遺伝子間の「重感染排除」の発見など寄生的ライフサイクルが解明された。 (3)制限修飾遺伝子の攻撃に対しての宿主細胞側の応答が解明され、「染色体切断による分離後細胞死」が実証された。 (4)細胞側の遺伝子相同組換え機構が染色体切断の修復によって細胞死に抵抗することが明らかになった。 (5)染色体上の制限修飾遺伝子とそれに連結した遺伝子を相同な遺伝子で置き換えようとしたところ、抵抗と大規模なゲノム再編が起きた。予期しない発見であった。 (6)このような制限修飾遺伝子による大規模ゲノム再編が自然界でも起きていることが、ピロリ菌のゲノム解読から示唆された。
