タンパク質の動的構造研究のためのX線溶液散乱解析システムの開発

研究概要

佐藤は、これまで使用してきた湾曲全反射ミラーによる入射X線を集光させるX線光学系に代わり、入射X線を「人工多層膜集光ミラー」で集光させる光学系をX線溶液散乱解析システムに組み込んだ。その結果、試料溶液に照射するX線強度を従来の光学系に比べて10倍以上向上させるとともに、入射X線の集光度も格段に向上させることに成功した。
また、片岡は、時分割測定の可能性を拡張するため、紫膜の光反応の時間分解X線散乱データに対して、特異値分解法を適用することにより、光吸収後に生成するシッフ塩基の脱プロトン化した中間体(M中間体)からの戻り反応において、2つの異なる構造を取ることを明らかにした。その結果、速い成分はMに、遅い成分はN中間体の特徴を示し、先に提唱したプロトンポンプの分子機構モデルの妥当性を支持する結果を得た。
一方、雨宮は、X線イメージインテンシファイヤーと冷却CCDを組み合わせた高感度2次元X線検出器を開発し、イメージングプレートまたは一次元位置敏感型ガス検出器と比較した。その結果、X線イメージインテンシファイヤー由来のコントラストの低下があることが判明したが、この問題を解決する方法を考案し、X線溶液散乱では、コントラストの低下の問題が完全に補正できることを示した。昨年度に行った補正ソフトの開発を併せて、本システムがX線溶液散乱実験において、高感度で時間分割測定に最適な検出器システムであることを明らかにした。さらに、蛍光体と縮小型光ファイバーをCCDに組み合わせたモジュールをモザイク状に並べたアレイ状CCD型X線検出器を製作し、3×6個のCCDをアレイ状に並べた有効面積236×236mmのCCD型X線検出器を製作した。
