p53遺伝子の変異した肝癌を標的とした遺伝子治療ベクターの開発

研究概要	日本人の肝癌では、癌抑制遺伝子p53が変異したものが30%前後あることが報告されている。本研究は、このp53遺伝子が変異したヒト肝癌を標的とした遺伝子治療ベクターの開発を目的としている。その為に、p53遺伝子が変異した細胞でのみ増殖できるようにしたアデノウイルス(Ad)と、p53遺伝子が変異した肝癌細胞でのみ高発現するHBV X遺伝子のプロモーター/エンハンサーを組合せ、その目的を達成させるべく研究を行った。アデノウイルスタイプ5(Ad5)ゲノムのサイズは約35kbであり、初期遺伝子と後期遺伝子を持ち、それぞれがゲノム複製とウイルス粒子形成に関与している。Ad5ベクターについては、E3或いはE1又はE4領域に欠損変異を持ったAd5 DNAをヒト293細胞にトランスフェクションして作成した。我々の研究から、HBVのX遺伝子のM4プロモーターは、p53遺伝子が正常な肝細胞では殆ど発現せず、p53遺伝子が変異した肝細胞でのみ著しく発現することが分かった。このM4プロモーターの下流に細胞のアポトーシスに関係している遺伝子を挿入して発現させれば、p53遺伝子の変異した肝癌細胞がアポトーシスに至ることが期待される。そこで、このM4プロモーターを用いた転写カセットを作成し、この転写カセットをAd5 DNA中のE3或いはE1領域に挿入することを試みた。現在、p53遺伝子が正常の細胞(HepG2やNIH3T3)およびp53遺伝子が変異した細胞(HuH7)を用いた感染実験を行っている。
