研究概要

日本人の肝癌では、癌抑制遺伝子p53が変異したものが30%前後ある。本研究では、p53遺伝子が変異したヒト肝癌を標的とした遺伝子治療ベクター開発を目的とした。P53変異細胞で増殖できるように操作したアデノウィルス(Ad)にp53変異肝細胞でのみ特異的に高発現するプロモーター/エンハンサーの下流に細胞死に関連した遺伝子を挿入し、肝臓細胞への指向性を利用するものである。
肝臓細胞で特異的に発現が調節されるB型肝炎ウィルス(HBV)のX遺伝子のM4プロモーターは、p53遺伝子が正常な細胞では殆ど発現せず、p53変異細胞でのみ著しく発現する。そこで、このM4プロモーターの下流に例えば細胞死に関係している遺伝子を挿入し、この転写カセットをAd5ベクター中に挿入すれば、p53変異肝癌の遺伝子治療に極めて有効な方法が開発されることが期待される。
細胞死関連遺伝子として、まずHBVのX遺伝子を用いた。X蛋白質は転写活性化因子として知られているが、最近の我々の研究から、X蛋白質の中央領域が細胞死に特異的に関係することが明かになった。X遺伝子をAd5ベクターのE1領域に挿入し、p53変異肝癌の遺伝子治療用の組換えAd5ベクターを作製した。またC型肝炎ウィルス(HCV)のコア遺伝子の利用も検討したが、細胞死作用が弱いことが分かった。これと並行してKB細胞由来のIL-6(KB)がp53変異細胞の細胞死を強く誘導する事を明らかにしたので、これをM4プロモーターの下流に挿入した転写カセットを作製する事を試みている。
いずれの場合も、培養細胞を用いての組換えAd5ベクターの活性についてはまだ十分に確立しておらず、今後の課題である。しかし、Ad5ベクターそれ自体の安全性が現在では問題となっており、開発の意義を考え直す必要が生じている。
