研究概要

脳梗塞や脳痴呆の治療薬は多数存在しているが、有用なものはきわめて少なく、しかもそれらの作用機序も不明な点が多い。PACAPはラットの海馬の遅発性神経細胞死を有意に抑制することを著者らはすでに報告している。さらに、著者は、虚血による遅発性神経細胞死過程の細胞内シグナル伝達機構にMAPキナーゼが関与し、そのうちJNK/SAPK活性が虚血により亢進することを明らかにした(Neurosci Lett,1999)。またPACAPは、虚血負荷においてJNK/SAPKのリン酸化を抑制することを著者は明かにしている(Ann NY Acad Sci,1998)。さらにPACAPのラットの脳室内投与実験により、髄液のlL-6が有意に上昇すること、またこのIL-6は反応性グリア細胞で産生されることが明らかになり、PACAPは神経細胞あるいはグリア細胞に作用して神経細胞死を抑制する可能性の高いことが明らかになった。著者は、PACAPによる神経細胞死防御を直接証明するために、PACAP レセプターのトランスジェニックマウスを作成し、虚血実験を行うことを計画した。最近ようやくその遺伝子改変マウスの作成に成功し、現在この遺伝子過剰マウスを繁殖させている。近日中にはこの遺伝子改変動物をもちいて虚血実験を行い、PACAPによる遅発性神経細胞死抑制の実体を明らかにする予定である。
