骨髄腫由来の異常血漿タンパク質の高感度・高分離能分析法の開発に関する研究

研究概要	疾患に由来する異常免疫グロブリン分子を高性能に分析できる方法を開発することを目的として、キャピラリー等電点電気泳動によるタンパク質の分析条件を検討した。まず、キャピラリー等電点電気泳動でタンパク質をそれぞれの等電点に収束させたのち、キャピラリーの端近くに設置した紫外吸収検出器の位置まで、タンパク質を移動させる条件を新規に考案し、タンパク質を等電点分離の時の相対位置を保ったままで分離できた。その他分析条件を詳細に検討することにより、ヒト血漿タンパク質を50分程度の分析時間中に約60の紫外吸収ピークとして分離できるという、高分離能かつ高性能な分析方法を確立した。この電気泳動条件は全自動のキャピラリー電気泳動装置にそのまま設定することができ、多数の試料の連続分析も可能となった。こうして決定した条件を用いて、従来法のセルロースアセテート膜電気泳動で骨髄腫と推定された患者血清試料の分析に適用したところ、すべての試料についてIgGが正常レベルの10倍から50倍高い紫外吸収ピークはとして検出できた。また、これらのIgGは腫瘍化した細胞が産生するものであるため、狭い等電点範囲に集中していることも同時に検出できた。これらの結果は、骨髄腫由来の異常血漿タンパク質の高感度、高分離能分析法としてキャピラリー等電点電気泳動がきわめて有効であることを示すものである。
