板鰓類の視床下部-下垂体系とゴナドトロピン分泌調節

研究概要	軟骨魚類は無顎類から四足動物への進化を探る上できわめて重要な動物群である。しかし軟骨魚類はシルリア紀に出現して以来、独自の進化を遂げてきたため、視床下部-下垂体系についても他の綱の動物と異なった特殊な点を色々ともっている。とりわけ、板鰓類の多くは体内受精をし、卵生から胎生まで生殖様式も多様で高度に発達していることを考えると、視床下部の直接支配の及ばない腹葉にゴナドトロピン活性があることは、きわめて興味深い。本研究では、まず(1)板鰓類の下垂体における腺下垂体ホルモンの分布を明らかにし、ついで(2)視床下部のゴナドトロピン放出ホルモン(GnRH)がどのような経路で腹葉に至るかを明らかにするとともに、(3)腹葉からゴナドトロピンを単離し、(4)RIA系を確立することをめざした。その結果、(1)の腺下垂体ホルモンの分布に関する研究では、成長ホルモン細胞やソマトラクチン細胞の分布を明らかにするなど、充分な成果をあげることができた。しかし(2)以下については、本研究期間内に充分な成果をあげることができなかった。一番の誤算は、サメの腹葉からゴナドトロピンを単離できなかったことがあげられる。夾雑物を最小限にするため腹葉が大きく発達した性成熟個体のみを集めて単離を進めるべきであったことや、抗体プローブのさらなる工夫をすべきであったことなど反省点も多い。一方、今回の免疫組織化学の結果が示しているように、板鰓類ではゴナドトロピンとサイロトロピンが分離しておらず、両者の祖先型が存在している可能性もある。さらに、GnRHニューロンの神経繊維も前脳には多数分布しているものの視床下部にはほとんど分布していないことなど、GnRHの放出経路については、謎が深まるばかりであった。一方、本研究と平行して研究が進められた円口類や原索動物では、当初の予想以上にめざましい成果を得ることができた。それらの成果もあわせて報告する。
