魚類卵の卵膜硬化カスケード

研究概要	メダカやニジマス等の魚類卵の卵膜硬化は、卵膜構成タンパク質の金属プロテアーゼによる限定分解と、トランスグルタミナーゼ(TGase)による高分子化(重合)反応の2つの反応が関与する。このカスケード系を理解するために、交付申請書に記載した3つの具体的な研究を行なった。その成果を交付申請書に従って以下に記載する。 (1)ニジマス未受精卵の単離卵膜の生理食塩水抽出液中にTGase活性が見られることはすでに分かっていた。この抽出液を出発材料として各種のカラムクロマトグラフィーを用いて2種類のTGase(P1、P2)を精製し、生化学的・酵素学的な性質を調べた。一般に知られているTGaseとほぼ同様の性質を持っていたが、P1 TGaseは特徴的で低イオン強度でのみその活性が発現するものであった。これは、卵が淡水中で受精反応を行ない、淡水中で速やかな卵膜硬化反応を進行させることを考慮すると、好都合な性質である。この成果の一部はすでに発表されている(Ha and Iuchi,1997;Ha and Iuchi、1997)。 (2)メダカ卵の卵膜硬化の過程で金属プロテアーゼによる構成タンパク質の限定分解が起こることはすでに報告した(Ha and Iuchi,1996)。ニジマス卵においても同様に金属プロテアーゼが関与するという事実が判明した。今後その精製と性格付けを試みたい。 (3)新しい発見があった。それは、卵付活後TGaseがある種のプロテアーゼにより限定分解され、同時にその活性が上昇するらしいことである。これは、上記のTGase、金属プロテアーゼの他に卵膜硬化カスケードの新しい成員が発見されたことを意味し、今後、このプロテアーゼの精製を試み、これら成員間の相互作用を調べる予定である。
