微生物由来ラクトン環開裂酵素における立体選択性制御機構の解明

研究概要	糸状菌Fusarium oxysporumの生産するD-パントラクトンを立体選択的に開環加水分解するラクトナーゼとバクテリアBrevibacterium protophormiaeの生産するL-パンラクトンを立体選択的に開環加水分解するラクトナーゼについて、構造および機能に関する解析を行い、反応機構・立体選択性制御機構の解明を試みた。精製したF.oxysporumのラクトン環加水分解酵素をプロテアーゼ消化し、それにより得られたペプチドのアミノ酸配列の決定を行った。このデータをもとにして作成したプローブを用いて、F.oxysporumより本酵素をコードする遺伝子のクローニングに成功し、本酵素のアミノ酸1次構造の決定を行うことができた。また、F.oxysporumより得られたラクトン環加水分解酵素標品を用いて、X線結晶構造解析に適した結晶を得るため、ハンギングドロップ法または透析法により結晶化条件の検討を行った。沈殿剤として、ポリエチレングリコール4000が効果的であり、構造解析により適した大きさ・質を有する結晶を調製することができた。また、B.protophormiaeよりラクトン環加水分解酵素の単離を行い、諸性質の解明を行った。上記と同様にプロテアーゼ消化による内部ペプチドの取得およびアミノ酸配列の決定に成功した。これらのアミノ酸配列のデータベースとの比較により、F.oxysporumのラクトナーゼおよびB.protophormiaeのラクトナーゼはともに新規な酵素であることが判明した。
