研究概要	本碩究は慢性肉芽腫症(CGD)を中心とした遺伝性白血球機能障害の情報伝達から見た解析と、それを通じて好中球の殺菌能として重要なNADPHオキシダーゼの活性化機構を明らかにすることを目的とした。本研究で明らかに出来た事として、次のようなものである。第一に正常好中球のNADPHオキシダーゼ活性化と貪食機構に関してp38MAPキナーゼという、最近発見された新しいキナーゼの関与を明らかにした。第二に好中球の活性酸素生成が欠損しているヒト慢性肉芽腫症(CGD)の細胞を用いた研究で、NADPHオーキシダーゼコンポーネントであるp47phox発現ベクターを発現させることでNADPHオキシダーゼ活性を回復させる事に成功した。これは遺伝子治療の基礎的研究として意義があると考えられる。またこのP47phoxの部位特異的突然変異法を用いた詳細な研究で、P47phoxの11個のセリン残基のうちS303,S304,S359,S370のリン酸化がNADPHオキシダーゼ活性化の引き金になること、スピンラベル法を用いた研究でp47phoxがリン酸化する際に、そのC末端領域の構造変化が起きることを明らかにした。第三にCGDと類似の活性酸素生成能が欠損しているウシ癲粒球粘着不全症(BLAD)の好中球は19Gや補体受容体刺激による活性酸素生成能は著しく低下しているが、その情報伝達系にPKCが関係するNADPHオキシダーゼ活性化システムの寄与が大きくなる(up-regulation)ことを明らかにした。ホスファチヂルイノシトール3キナーゼ、p38MAPKあるいはチロシンキナーゼが関与する機構については寄与の程度は正常なものと変化は無かった。第四にBLADのウシに正常ウシの骨髄の移植を試みたところ、28カ月にわたりCD18の好中球での発現が観察され、臨床的症状の改善が見られた。これは、今後のBLADへの骨髄移植の適用と言う点で意義があると考えられる。第五にこの様な血球細胞からの活性酸素は生体系では、炎症、虚血時の傷害と深く結びついて考えられているが、その防護機構として、活性酸素に曝された細胞をその後、Troloxやカテキンなどの抗酸化剤で処理することにより軽減出来ることを細胞レベルとスナネズミの脳虚血モデルで見いだした。これは活性酸素傷害に対する新たな防護法の確立と言う点で意義がある知見である。以上のようなNADPHオキシダーゼの活性化機構をCGDならびBLADにからの細胞を用いることで明らかにすることができた。さらに欠損酵素のコンポーネントの発現、骨髄移植、酸化傷害からの抗酸化剤による防護法など、将来への治療という点でも意義のある成果が上げられた。
