研究概要	(1)生物膜を反応場(高粘度環境)とする酵素機能の基盤解析と新規解発: 生物膜を構成する複合脂質が酵素分子の構造形成と触媒活性にどう関わっているかを精査する目的で、従来蓄積された研究データと比較検討可能なモデル系(高粘度環境)を設定し、ureaの共存が酵素(グルコアミラーゼ)機能及び立体構造に及ぼす効果を速度論的(定常状態、遷移相)、平衡論的に調べ、反応機構を解析した。その結果、立体構造の大きな変化は観られないが、二段回機構の第1素過程(非生産中間体)を採り易くなることが知られた。 (2)標的蛋白質の採取とキャラクタリゼーション: 3種の標的(膜)蛋白質・酵素の採取・精製を行って分子量、分子構築、生物・生命機能を検討した。1)酵母Kluyveromyces lactisから採取したβ-ガラクトシダーゼは2つの単量体(分子量120、000)で構成され、反応環境(粘度)が立体構造及び触媒活性に強く影響すると知られた。2)ウシ脳を各種処理し、コール酸ナトリウム溶性画分から電気泳動的に単一な標品(膜酵素β-グルコセレブロシダーゼ)を得、二糖類を基質として機能解析した結果、ウンベルフェリル β-グルコシドに特異性を持つと判った。3)ウシ茸状乳頭(味蕾)を顕微鏡(本研究費補助金により購入)下に採集、Triton-X可溶画分を植物Gymnemaa sylvestreの葉より採取したギムネマ酸を活基とするアフィニティーカラムに依り、甘味剤受容蛋白の分別・精製に成功した。分子量は34、000で、甘味二糖に対する触媒(水解、転移)活性は認めなかった。 (3)ヴェシクル(リポソーム)の調製による膜酵素の再構築: ホスファチジルコリンを構成材としてヴェシクル(MLV)を調製し、Asp.niger起源の酵素粗標品から精製したβ-グルコシダーゼを用いて、膜酵素への再構築を試み、各種糖基質に対する触媒活性を指標としてヴェシクルの構築確認と機能分析を行った。
