下垂体前葉細胞の分泌機能に対するミオシンおよびミオシン軽鎖キナーゼの役割

研究概要	昨年度までの研究によってマウスの下垂体前葉細胞の分泌果粒にアクト・ミオシンがassociateしている可能性が示唆されていたが、これをさらに確かめるために次の実験を行った。 1. 下垂体前葉細胞をTriton Xでpermeabilizeし、heavy meromyosinに浸漬した。これを超薄切片にして電子顕微鏡で観察したところ、heavy meromyosinで修飾されたF-アクチンが分泌果粒膜に結合していた。その結合様式は2通りであり、F-アクチンの一端が直接分泌果粒膜に結合する場合と、F-アクチンと分泌果粒が短い架橋によってside-to-sideに結合する場合とがあった。 2. 下垂体前葉細胞を急速凍結エッチングレプリカ法で電顕観察すると、5nm周期のstripesで同定されるF-アクチンが分泌果粒に結合ていた。その結合様式は超薄切片法で観察した場合と同様であった。 3. F.アクチンと分泌果粒をside-to-sideに架橋する構造がミオシンではないかと推測されるので、次にアクチンとミオシンの二重免疫電顕法を試みた。下垂体前葉をLowicrylに包埋し、その超薄切片を抗アクチン抗体と抗ミオシン軽鎖抗体で免疫標識して電顕で観察した。分泌果粒の近傍でアクチンとミオシンが共存し、さらに果粒膜とアクチンの間にミオシンが局在する部位が検出された。 4. 以上の結果から、下垂体前葉細胞の分泌果粒にアクト・ミオシン系が結合していることが確実となったが、さらにミオシンの機能を制御するミオシン軽鎖キナーゼの局在を免疫電顕法で調べた。その結果、分泌果粒膜に近接してミオシン軽鎖キナーゼが局在していることがわかった。これらのことから、下垂体前葉細胞の分泌果粒の輸送にアクト・ミオシン系が関与していることが強く示唆された。今後は、分泌果粒の輸送に関与する微細管・キネシン系とアクト・ミオシン系の2つの異なるシステムがどのように協調して果粒を運んでいるかを解明したい。
