下垂体前葉細胞の分泌機能に対するミオシンおよびミオシン軽鎖キナーゼの役割

研究概要

アクト・ミオシン系による分泌顆粒の運動がプロテインキナーゼによってどのように調節されているかを検討するために、分泌調整機構が詳しく解析されているMIN6細胞(膵B細胞由来)をモデル系として解析を行った。
1.ミオシン軽鎖キナーゼ(MLCK)はミオシン軽鎖を特異的にリン酸化することによって、アクチンとミオシンの相互作用を制御することが知られている。MIN6細胞における分泌顆粒の分布は、cytoplasmic type、intermediate type、peripheral typeの3つのパターンが見られたが、いずれのタイプでもMLCKと分泌顆粒の分布にかなりのオーバーラップが認められた。
2.さらに免疫電顕法によって、分泌顆粒膜の外側にMLCKの局在が証明された。したがって、アクト・ミオシン系による分泌顆粒の運動はMLCKによる調節を受けていると考えられる。
3.protein kinase A(PKA)とprotein kinase C(PKC)によるリン酸化の影響を知るために、adenylate cyclaseのactivatorであるforskolinとPKCのactivatorであるTPAの効果を調べた。TPA単独では分泌顆粒とMLCKの分布はcytoplasmic typeからperipheral typeへ移行したが、forskolin単独では顆粒とMLCKの分布に変化はなかった。生きているMIN6細胞での分泌顆粒の動きを位相差顕微鏡で観察すると、forskolinによって顆粒の運動は活発になったが、TPAでは変化がなかった。
4.forskolinとTPAを同時に与えると、顆粒とMLCKの分布はcytoplasmic typeからperipheral typeへシフトし、顆粒の運動も活発になった。
これらの結果から、MLCKだけでなくPKAとPKCも分泌顆粒の細胞内動態に影響を及ぼすことが示唆された。
