微小循環系(リンパ管・血管)の臓器内分布と新生に関する組織化学的研究

研究概要	1 正常および病理組織における微小脈管(リンパ管・血管)の確定のための酵素組織化学法と免疫組織化学法の適用の可能性(対比) 酵素組織化学法:5'-nucleotidase(5'-Nase)染色によりリンパ管を、alkaline phosphatase(ALPase)染色により動脈およびdipeptidyl aminopeptidase IV(DAP)染色により静脈の三者の染別(二重・三重染色)をヒト及び実験動物(マウス・ラット・ハムスター・サル等)の種々の組織や器官の切片や伸展試料(全載標本)で試み、良好な結果を得た。さらに、腸管壁については、粘膜層や筋層を注意深く剥離し、薄層の伸展試料を5'-Naseとacetylecholine esteraseの二重染色し、リンパ管と神経の光顕像とSEM反射電子像(硝酸銀処理)を観察し、両者の分布状況を明らかにした。免疫組織化学法:基底膜構成物質であるIV型コラーゲンやラミニンに対する抗体を用いて、これらの免疫染色により、血管も染別・同定した。また、nitric oxide synthesis(NOS)の分布も調べた。さらに、最近入手し得た抗ヒト胸管抗体を、ヒト胆褒組織を用いて検討(免疫染色)し、特異性を調べた。尚、現在上記両方法を併用することにより、両試料(切片・伸展)ともにより効果的な観察法を検索中である。 2 リンパ管の再生・新生実験 1)ラットの下腿部のリンパ管を切断、あるいは腹部胸管を結紮・切断し、一定期間後(1〜2週目)の再生時に局部の両脈管の増殖・新生状況を調べた。特に、新生リンパ管の5'-Nase酵素活性は対照群のそれらと比較して高かった。 2)腸間膜の酵素染色・免疫染色による両脈管の内皮細胞の増殖(出芽)の検索や培養リンパ管内皮細胞の細胞化学的検索は目下進行・検討中である。
