研究概要

【先体内物質の輸送経路】細胞内小胞輸送の阻害剤であるブレフェルジンを添加して、精子細胞の粗面小胞体からゴルジ装置への輸送過程およびゴルジ装置から先体への輸送と先体形成が障害されることを証明した(Tanii et al.(1998)J Electron Microsc,47;161.)。【精巣上体内成熟過程におけるacrin 1(MN7=90K)の修飾】先体内物質であるacrin 1は、精巣上体内成熟に伴う形態変化に伴って、先体内で局在変化することを証明した(Yoshinaga et al.(1998)Cell Tiss Res,292;427.)。【Equatorin(MN9=48K)の機能】Equatorinは、先体反応中に一部放出されるが、多くは精子が囲卵腔に到達するまで残存すること、抗equatorin抗体存在下の体外受精実験により、精子は卵子に進入できずに囲卵腔に蓄積し卵子から表層粒が放出されないこと、そして前核形成や2細胞期への発生が阻害されることを証明した。これらの事実から、equatorinが精子-卵子細胞膜融合過程に必須な分子であることが判明した(Toshimori et al.(1998)Biol Reprod,59;22.)。【Acrin 1とAcrin 2(MC41=165K)の機能】Acrin 1は先体反応に必要であり、acrin 2は先体反応後から透明帯通過までに必要な分子であることを証明した(Saxena et al.(1999)J Reprod Fert,accepted)。【Acrin 3(MC101=155K)の機能】Acrin 3は卵子との膜融合に関与することが示唆された。【MC31抗原(分子量26-35K)のクローニング】cDNA検索の結果、MC31抗原はラットCE9に高いホモロジーを示し、イムノグロブリン・ファミリータンパクであることが判明した。 【総説】精巣に由来する分子の成熟と受精過程における機能について総説した(Toshimori K(1998)Cell Tissue Res,293;177.)。
