食塩負荷環境適応における腎交感神経系と神経ペプチド系とのクロストーク

研究概要	未だ病因の不明な本態性高血圧症患者の一部は食塩負荷で昇圧しやすく食塩感受性群と呼ばれている。この規定因子には遺伝因子とともに種々の要因が想定されているが、腎交感神経系を介する腎臓からのナトリウムの排泄異常が関わっているのではないかという作業仮説のもと、これまで覚醒下のDahl食塩感受性ラットを用いて実験を行ってきた。高食塩食の長期間摂取に対する生体の長期的適応機構に、はたして神経性調節系がどの程度関わっているかについては論議がある。血圧変化のセンサーである圧受容器からの求心性線維を切断してもその効果は一時的なもので永続しない。また圧受容器は持続的刺激に対して速やかに順応してしまう。しかし液性調節系のバゾプレッシン系やレニン・アンギオテンシン系の応答にはそのような順応現象は認められていない。また最近自律神経活動はこれらペプチドによって修飾されることが明らかにされた。したがって食塩負荷に対する適応に自律神経系とともに内分泌系とクロストークが主要な働きをしていることが考えられる。今回、中枢性高張食塩水負荷に対してあらわれる腎交感神経抑制反応について、圧受容器からの入力とともにバゾプレッシンなどホルモンが如何に関わっているか、そしてホルモンの受容部位と想定されている脳室周囲器官の一つである最後野(AP)の関与について調べた。 1.覚醒・自由行動下Sprague-Dawleyラットにおいて、脳室内に高張食塩水を注入すると血圧の上昇を伴って腎交感神経活動が用量依存性に抑制された。 2.この腎交感神経活動の抑制反応には圧受容器からの入力と、V_1レセプターを介するバゾプレッシン系とクロストークの関与が示唆された。 3.食塩負荷後APにはFos蛋白陽性ニューロンが認められたが、AP破壊によって食塩負荷に対する腎交感神経応答は影響を受けなかった。
