神経型一酸化窒素合成酵素のカルシウム情報伝達系における活性制御の解析

研究概要	研究目的:本研究では新たなるカルシウム情報ネットワークの解明のためカルモデュリン(CaM)により活性化される酵素のなかで神経型NO合成酵素(neuronal NO synthase:nNOS)を介した中枢神経機能を分子レベルで解明することを目的とし、以下の研究を行った。研究項目:1. nNOSの活性制御の解析。[(1)nNOSのCaM結合に本質的なアミノ酸の同定。(2)nNOSの構造と細胞内局在について。(3)神経細胞におけるnNOSと誘導型NOSによるNO産生調節について。] 2. nNOS選択的阻害剤の創製とその構造活性相関の検討。3.新たなるCa^<2+>/CaM受容蛋白の検索。結果及び考察:1.(1)の結果を踏まえ非活性型nNOSミュータントを得ることができた。1.(2)それを分子生物学的、細胞生物学的に用いnNOSの膜分画局在がそのCaM結合部位によることを示唆した。1.(3)同一細胞にnNOSと誘導型NOS(iNOS)が存在する場合、カルシウム感受性を有さないiNOSがnNOSとの間で2量体を形成し、そのいわゆるハイブリッドダイマーがカルシウム感受性を獲得することを示した。2.NOSの中でnNOSにのみ選択的に阻害作用を示す化合物を創製し、その構造活性相関を明らかにして分子薬理学的道具としての有用性を示した。3.カルシウム情報ネットワークに存在する新規機能蛋白として、CaMキナーゼIの新しいアイソフォームのクローニングを通してアイソフォーム特異的なOMキナーゼIの活性化メカニズムを発見した。また、中枢神経に発現が多いカルシウム結合蛋白、ニューロカルシンの新しいアイソフォームのクローニングとその生化学的性格の分析を行った。今後の研究課題:現在までに得られた知見を更に発展させ、カルシウムシグナルにおけるNOSとCaMキナーゼ、或いはカルシウム結合蛋白とのクロストークによる活性制御機構解析の研究を展開させたい。
