ヒト血小板接着・凝集におけるインテグリンシグナルの解析

研究概要	血小板活性化におけるリガンドのGPIIb/IIIa結合により引き起こされる細胞骨格の再構築、血小板凝集、血餅退縮に至るoutside-inシグナルを明らかにするために、GPIIb/IIIaに対する特異抗体やリガンド結合部位に対するアンタゴニスト(RGDS)を用いて、チロシンリン酸化、イノシトールリン脂質代謝の影響を検討しoutside-inシグナルの実体を解明することを試みた。トロンビン刺激により血小板が凝集すると細胞骨格の再構築が起こり、PI-PLCが細胞骨格に移行する。この移行はGPIIb/IIIaのアンタゴニストRGDSの処理、および特異的抗体により阻害される。したがって、リガンドのGPIIb/IIIa結合によるインテグリンを介するシグナルにPI-PLCの活性化が関与することが示唆された。また、PLCβ3のサイトソル型が特異的に細胞骨格に移行し、このタイプはGβγによって活性化されることが確認された。フィブリノーゲンをコートしたプレートに血小板を蒔き、フィブリノーゲン結合による全チロシンリン酸化を検討したところ、FAKのチロシンリン酸化が増加していた。ヒト血小板のモデル細胞として巨核芽球性白血病細胞Meg-01を用い、血小板への分化過程における上記情報伝達因子の発現様態を検索し、接着凝集シグナル因子を同定することを試みた。まず、分化誘導因子としてPMAによる巨核球への分化誘導を検討した。分化マーカーとしてGPIIb/IIIaの発現増加を調べ、分化が誘導されていることを確認した。本細胞は血小板に存在するものと同じPLCアイソザイムを含み、分化誘導過程におけるPLC,PKCアイソザムの量的変動を検討したところ、PKCβIIとPLCγ2の発現増加が観察された。今後、これらの増加とGPIIb/IIIaシグナルの関連性を検討する。
