インスリン分泌顆粒の運動制御機序の解明

研究概要

膵B細胞におけるカルシウムシグナリングの情報受容蛋白であるミオシンについては、生化学と形態学的アプローチの結果、ミオシンが細胞内の分泌顆粒状に局在し、その軽鎖のリン酸化が細胞内遊離Ca^<2+>とカルモジュリンに依存してリン酸化を受け、細胞内の分泌顆粒は「貯蔵プール」から「準備状態」に運ばれることを実証し、さらにアセチルコリンがこの情報伝達系を介してインスリン分泌を増加させることを突き止めた。ミオシン軽鎖のリン酸化は、膵B細胞に特微的なCa^<2+>要求性を有しており、この分子機序を現在検討中である。もう一方のモーター蛋白質であるキネシンについても、その微細形態を検討中である。さらに私達の実験により、ミオシンには内皮細胞の細胞内Ca^<2+>濃度調節や胚細胞の発生過程で多彩な役割を果たすことが次々と明らかになり、現在このカルシウム結合蛋白の多彩な機能の解明に取り組んでいる。
一方カルサイクリンについては、これまで困難であった齧歯類のカルサイクリンに反応する特異抗体の作製に成功し、これを用いてその分布を検討するほか、ストレプトリシンOを用いた透過性細胞を用いて分泌への影響を調べている。さらにカルサイクリンの特異的結合蛋白質であり、カルシウム/リン脂質結合するアネキシンの役割も透過性細胞と新たに作製した抗アネキシンXI抗体を用いて検討中である。また、別のアイソフォームであるアネキシンIIの細胞内分布を調べ、この蛋白質が開口放出における顆粒と細胞膜の癒合ばかりでなく、Ca^<2+>依存性の細胞内膜癒合現象に広く関与していることが示唆された。
