インスリン分泌顆粒の運動制御機序の解明

研究概要	膵B細胞における顆粒運動はアセチルコリンによって活性化される。この活性化において、アセチルコリン刺激による細胞内Ca^<2+>遊離が、Ca^<2+>/カルモジュリン依存性にミオシン軽鎖リン酸化酵素を活性化し、インスリン顆粒のモーター蛋白質であるミオシンを活性化し、分泌顆粒を分泌可能なプールまで運ぶという、細胞内情報伝達の仕組みを私たちは明らかにした。現在は最も重要な分泌刺激であるグルコースによる顆粒運動活性化機構の解明にあたるとともに、ある種の刺激条件下でミオシン軽鎖リン酸化酵素の細胞内分布が変化することを見いだし、これとインスリン分泌調節との関係を調べている。一方力ルサイクリンについては、作成した齧歯類のカルサイクリンに反応する特異抗体を用いて細胞内分布を調べた。膵島では、カルサイクリンは細胞質に不均一に分布したが、培養膵B細胞であるMIN6では、そのシグナルは非常に弱かった。一方、カルサイクリンの特異的結合蛋白質であるアネキシンXIに対する抗体による免疫染色では、膵島細胞およびMIN6細胞において細胞質中に顆粒状に免疫陽性像が得られ、その一部はインスリン顆粒と共存していた。この結果はアネキシンXIが、顆粒のトラフィックやフージョンに働いている可能性を示している。現在、アネキシンXIおよびその抗体の分泌に対する影響を、透過性細胞を用いて調べている。
