Ca^<2+>・リン脂質結合領域(C2様領域)を有する新しい蛋白質の機能と作用機構

研究概要

神経伝達物質の放出には、プレシナプス内へのCa^<2+>の流入が必要であるが、私共が見い出したRabphilin-3AやDoc2は、リン脂質依存性にCa^<2+>と結合するC2様領域を有することから、私共は、これらのタンパク質がCa^<2+>センサーとして神経伝達物質の放出を制御していると考えている。本研究では、Rabphilin-3AやDoc2の機能と作用機構を明らかにすることによって、神経伝達物質の放出機構の全貌を分子レベルで解明することを目的としており、本年度は以下の成果を得た。
1)Rabphilin-3Aの機能と作用機構 : ヤリイカの巨大シナプスの系を用いてRabphilin-3Aが実際に神経伝達物質の放出を制御していることを明らかにした。さらに、その際、Rabphilin-3Aは、シナプス小胞のプレシナプス膜へのドッキングと神経伝達物質の放出後のシナプス小胞の取り込みに機能していることが明らかになった。
2)Doc2の機能と作用機構 : シナプス小胞に局在するDoc2がプレシナプス膜に局在するMunc13に結合することを見い出した。さらに、Doc2のMunc13結合領域のペプチドを用いた細胞生物学的解析により、Doc2とMunc13の結合が神経伝達物質の放出において機能していることを明らかにした。また、Doc2のノックアウトマウスを作製して神経生理学的解析を行ない、Doc2がシナプス小胞のreadily releasable poolからの放出を阻害すると共に、シナプス小胞のreserve poolからreadily releasable poolへの移動を促進するのに機能していること、さらには、海馬のシャッファー側枝のシナプスにおけるLong-Term Potentiationにも関与していることを明らかにした。
このように、本年度の研究は予想以上に進展し、当初の目的はほぼ達成することができた。
