研究概要

肺サーファクタント蛋白質SP-AおよびSP-Dは、C末端側に糖鎖認識領域(CRD)を有するC型レクチンのコレクチン・サブグループに属する。SP-AとSP-Dの脂質結合特異性は、特徴的で、SP-Aはジパルミトイルホスファチジルコリン(DPPC)に結合し、リン脂質リポソームの凝集を惹起する。SP-Aは肺胞II型細胞とも相互作用を有する。一方、SP-Dは、ホスファチジルイノシトール(PI)に特異的に結合する。コレクチンに属するマンノース結合蛋白質A(MBP-A)もPIに結合する。また、糖脂質では、SP-Aがガラクトシルセラミド(GalCer)に、SP-Dがグルコシルセラミド(GlcCer)に結合する。本研究は、CRD内におけるSP-AとSP-Dの脂質結合部位を検索し、肺胞II型細胞との相互作用におけるSP-Aの機能領域を明らかにすることを目的として、SP-A CRDの一部をMBP-A CRDあるいはSP-D CRDに置換したSP-A/MBP-Aキメラ分子及びSP-A/SP-Dキメラ分子をバキュロウィルス-昆虫細胞系で発現、作成してその機能を調べた。SP-A/MBP-Aキメラ分子の解析からMBP-ACRDのGlu185-Ala221領域はSP-ACRDのGlu195-Phe228領域にSP-A機能を損なうことなく代替できることが示された。また、SP-A/SP-DキメラではSP-ACRDのGlu195-Phe228をSP-DCRDのGlu321-Phe355に置換したキメラ分子をad3、さらに、SP-AのThr174-Phe228をSP-DのThr300-Phe355に、また、SP-AのCys128-Phe228をSP-DのCys266-Phe355に置換したキメラ分子を、それぞれ、ad4とad5としてその機能を解析した。adキメラ(ad3-5)はSP-Aの持つ脂質結合性と肺胞II型細胞に対する機能を失っていた。wtSP-Dと同様にキメラad5はPIとGlcCerに強く結合したが、ad4は、PIとのみ、結合し、GlcCerには、ほとんど結合しなかった。ad3はPIに弱い結合性を示したが、ClcCerには結合しなかった。以上の結果は、SP-Aの脂質、および、肺胞II型細胞との相互作用には、SP-A CRDのC末端側Glu195-Phe228の領域が必須であること、さらに、SP-DのPIとGlcCcerへの強い結合には、SP-DCRDのCys266-Phe355領域が必須であるが、PIとGlcCer結合の機序は異なっていることを示している。
