LIM・PDZ構造を持つLIMK2の機能解析と発生分化及び癌化に於ける役割の解明

研究概要	我々は蛋白-蛋白結合に関与するLIM・PDZドメインを持ち、肺癌細胞株の一部で発現増強が見られるLIMK2遺伝子を単離し、この遺伝子の各種組織の発生分化および癌化への関与について、主として固形腫瘍発症との関連を中心に検討を進めている。本年度の研究に於いては、転写開始点の違いにより生じるLIMK2遺伝子の2種のisoformであるLIMK2a・2bの組織特異的発現制御機構とその生物学的機能の解明を目指して、第一に正常組織及び種々の癌細胞株に於ける2a・2b isoformの発現量を定量的に解析した。その結果組織特異的な発現調節を確認すると共に、一部の癌細胞株で2b isoformの異常な発現増強が見られることを見い出した。又、2a・2b各々の遺伝子構造及び転写開始点の確定・特異的プロモーター領域の単離を終了し、プロモーター活性を解析し発現制御機構の解明を進めた。現在2a・2b isoformの発現比率と、単離した各々のプロモーター活性が相関することを確認し、その転写活性領域の同定を進めており、発現制御機構の解明を目指している。又、テトラサイクリンレプレッサーの系を用いてLIMK2a・2b各々をヒト正常気管支上皮細胞株へ遺伝子導入し、各々を発現誘導し得る細胞株を樹立し、LIMK2遺伝子の細胞生物学的機能の解析を開始した。増殖能の変化・分化誘導等を示唆する初期的結果が得られており、更に解析を進めている。一方、LIMK2のLIMドメインに結合する遺伝子の候補のひとつとしてヒトLdb2遺伝子を単離し、LIMK2遺伝子の機能との関連を中心に検討を始めている。
