消化管癌におけるサイクリンEの癌遺伝子としての役割の解明

研究概要

1. サイクリンE発現ベクターの線維芽細胞株への導入
(1) 前年度作成した恒常的サイクリンE発現ベクターを、リポフェクチン法によりNIH/3T3マウス線維芽細胞株に導入した。導入後、ネオマイシン誘導体G418にて選択を行い、pCDNA3空ベクターのみの導入株を含め、各細胞株につき10クローンを回収した。
(2) サイクリンE発現ベクター導入NIH/3T3マウス線維芽細胞株において、DNAレベルならびに蛋白レベルでサイクリンEcDNAの組込みならびに発現を確認した。
(3) サイクリンE発現ベクター導入NIH/3T3マウス線維芽細胞株とpCDNA3空ベクター導入株の平皿培養下での増殖態度の差を観察したところ、両群の間に有意の増殖速度の差を認めることは出来なかった。また、サイクリンE発現ベクター導入株において形質転換フォーカスの形成はみられなかった。
2. サイクリンEアンチセンスベクターの胃癌細胞への導入
(1) 前年度作成したサイクリンEアンチセンスベクターを、リポフェクチン法によりサイクリンE過剰発現株TMK-1、MKN-7胃癌株、サイクリンE低発現株MKN-28、MKN-74胃癌株に導入した。導入後、ネオマイシン誘導体G418にて選択を行い、ベクターのみの導入株を含め、各細胞株につき10クローンを回収した。
(2)(1)で得られたサイクリンEアンチセンスベクター導入胃癌細胞株とpCDNA3空ベクター導入株の平皿培養下での増殖態度の差を観察したところ、両群の間に有意の増殖速度の差を認めることは出来なかった。
以上より、今回の研究ではサイクリンEが癌遺伝子として作用する直接の証拠を得ることは出来なかった。
