角化細胞の自己分泌増殖抑制因子とがん化に関する研究

研究概要	1. TGF-β特異型SMAD,Smad2とSmad3の機能解析:Smad2Δexon3というスプライス変異体が存在した。Smad3によって発現が強く誘導されるp3TP-luxをレポーターとした場合、この変異体はSmad3と同等の発現誘導能を示した。さらにこのプロモーター領域にはSmad3は結合できるがSmad2は結合できない配列があるが、Smad2Δexon3はこの配列に結合能し、exon3によってコードされた30アミノ酸がSmad2とSmad3のDNA結合能の違いを決めていた.(文献1)。また、角化細胞株HaCaTにおいて、TGF-βはSmad2とSmad3の両者を強く活性化したが、アクチビンは主にSmad3を活性化した(文献3)。 2. 変異SMADによるTGF-βシグナルの抑制:大腸癌でSmad2に見いだされた変異と相同な変異をSmad3に導入したSmad3D407EはTGF-βシグナルに優性抑制作用を示した。この変異Smad3を角化細胞株HaCaTに発現させると、TGF-βによる増殖抑制作用に抵抗性となった(文献2)。さらに、TGF-βに抵抗性を獲得した角化細胞は浸潤性増殖能を獲得していた。 3. がんにおけるTGF-βの異常:甲状腺乳頭癌は、免疫組織化学的検索によるTGF-βの染色性が高い症例に転移が多かった(文献4)。 4. TGF-βシグナルの標的遺伝子:TGF-βによる角化細胞の増殖抑制においてc-mycがSMADの標的遺伝子になっていた。また角化細胞におけるTGF-βの初期応答遺伝子のcDNA配列を決定した。この遺伝子は分子量19kDa、等電点10.9の新規塩基性核蛋白をコードしていた。 5. TGF-βシグナルの生理機能:ラット角膜の創傷治癒モデルにおいて、TGF-βが角化細胞の遊走に関与することが示唆された(文献5、6)。
