研究概要	黄色ブドウ球菌のロイコシジンおよびγヘモリジンは、二成分細胞崩壊毒素であり、ロイコシジンはLukSとLukF、γヘモリジンはHlg1とHlg2タンパク質から成る。我々はすでに、γヘモリジンの二成分が、ヒト赤血球の膜上でモル比1:1の割合で集合してリング状の膜孔(membrane pore)複合体を形成する事実を示した。本研究では、ロイコシジンの二成分が、ヒト多形核白血球およびウサギ赤血球の細胞膜においてモル比1:1の割合で集合してリング状の膜孔(membrane pore)複合体を形成する事実を明らかにした。ロイコシジンのリング状膜孔複合体を標的細胞膜から単離して、SDSポリアクリルアミド電気泳動および電子顕微鏡によって分析した。そに結果、ロイコシジンの膜孔複合体は、γヘモリジンの膜孔複合体と同様に、約200kDaの大きさであることが判明し、複合体はLukSとLukF成分を各々3分子含有していると考えられる。一方、ロイコシジンの膜孔複合体は、γヘモリジンの膜孔複合体と比較して構造的にやや不安定であり、細胞膜の可溶化処理によって4量体に相当する140kDaの毒素オリゴマーを生成した。また、ロイコシジン膜孔の実効内径は1.9-2.1nmであったが、この値は、γヘモリジンの膜孔内径(2.1-2.4nm)に比較すると僅かに小さい値であった。
