研究概要

これまでに我々は、動物感染モデルを用いてCryptococcus neoformansやPenicillium marneffeiに対する感染防御機構における細胞性免疫能の重要性、特にC.neoformansではIFN-_y、TNF-α、IL-12、IL-18を中心としたTh1関連サイトカインの重要性、マクロファージ殺菌機構における一酸化窒素(NO)、活性酸素の役割について明らかにしてきた。これらの知見に加えて本年度の研究では、IL-12とIL-18が相乗効果的にNK細胞及び_yδT細胞からのIFN-_y産生を誘導することでC.neoformans感染に対して防御効果を有すること、この作用にはTNF-αがマクロファージからのNO産生を増強することで関与していることを明らかにした。一方、我々はC.neoformans感染後肺内では炎症細胞の集積が極めて乏しく真菌が肺胞腔を埋め尽くしているような像を観察していたが、本年度の研究ではこの現象をフローサイトメトリを用いて確認するとともに、各種ケモカインの産生動態を解析することでケモカインの産生異常がこのような病理学的変化を起こしている可能性を示す結果が得られた。逆に、IL-12を投与すると著明な炎症反応が誘導され致死的感染から免れるが、これはケモカインの産生異常が是正されるためと考えられた。
一方我々は、host-parasite interactionの中で宿主側からだけでななく真菌から宿主への作用も存在し、C.neoformansやCandida albicansが活性化マクロファージからのNOの産生を抑制するという現象を見出してきたが、本年度の研究ではC.neoformansが同細胞からのIL-12産生を抑制しIL-10産生を軽度増加させることを報告した。これらのことは、host-parasite interactionを考える上で大変興味深い。また、P.marneffeiに対するマクロファージの殺菌機構として活性酸素よりもむしろNOがより重要であり、活性酸素はNOによる殺菌に対して調節的に働くことを示した。今後は、これらの知見をさらに拡げることによって、これら真菌の発症病態を解明していきたい。
