インターロイキン4によるIgE産生機構の解析

研究概要	(1) アレルギー疾患は多因子遺伝病で、その遺伝的素因はアトピーと呼ばれている。IgE産生誘導作用を持つIL-4、IL-13はアレルギー疾患において重要な役割を持っているが、これらのレセプターの共通のコンポーネントであるIL-4レセプターα鎖(IL-4Rα)がアトピーの原因遺伝子の一つであるかどうか検討した。その結果、1)アトピー性喘息患者において、IL-4Rαの50番目のアミノ酸におけるイソロイシンの出現頻度はバリンに比べて高かった。2)B細胞株にイソロイシン型のIL-4Rαを発現させると、バリン型を発現させた場合に比べてIL-4シグナルが増強していた。3)イソロイシン型の末梢単核球では、バリン型に比べてIL-4によるIgE産生が増大していた。このことから、IL-4Rαの50番目のアミノ酸におけるバリンからイソロイシン残基への置換はアトピーの原因となりうることを示しており、IL-4Rα遺伝子がアトピー原因遺伝子の一つであることが確認された。 (2) 転写因子であるSTAT6はIL-4のシグナルに重要であるが、IL-4Rα上のSTAT6活性化に必要な部位を決定した。さらに、この部位に結合する分子として、p47^<phox>を同定した。これは、IL-4の新しいシグナル伝達分子であると考えられた。 (3)IL-4の別のスプライシング産物として、エクソン2の部分が欠失しているIL-4δ2というものの存在が知られている。T細胞に対して、これ自身はIL-4のシグナルを伝達せず、IL-4の拮抗阻害剤として働くことが知られていた。それ以外の細胞に対するIL-4δ2の作用を解析したところ、B細胞、単球に対してもIL-4の拮抗阻害剤として働くことが判明し、IL-4δ2がアレルギーなどの治療薬として使用できる可能性を示した。
