エンドトキシン病態におけるIFN-γ誘導因子(IGIF)の役割に関する研究

研究概要	本研究では、IL-18の生理活性を、特に、そのエンドトキシン病態における役割に焦点を当てて検討した。第一に、IL-18(IGIF)のノックアウトマウスを作製し、野生型マウスと比較検討した。その結果、IL-18ノックアウトマウスはエンドトキシン肝障害にはならないが、ショック死に至りやすかった。このことから、IL-18はエンドトキシン肝障害に必須であることが判明した。また、IL-18欠損マウスのリンパ球は、野生型のそれと同程度にTh2細胞へ分化したが、Th1への分化に異常があった。また、IL-18欠損マウスは野生型とほぼ同数のNK細胞を保有するが、そのNK活性は低下していた。このように、免疫系の分化にもIL-18が大きく貢献していることが判明した。第二に、種々の細胞におけるIL-18受容体の発現を検討したところ、NK細胞は恒常的にIL-18受容体を発現しているが、ナイーブなT細胞はIL-12刺激を受けることにより本受容体を発現する事が判明した。また、IL-18の細胞内伝達機構もMyd88欠損マウスを用いて明らかにした。また、IL-18受容体ノックアウトマウスはIL-18刺激にまったく応答しないことから、IL-18受容体が、IL-18の主要な受容体であることが判明した。第三に、IL-18の分泌にIL-1β転換酵素(ICE)が重要な働きを果たすことを、ICEノックアウトマウスを用いて証明した。第4にICEを多量に発現するICEトランスジェニックマウスが恒常的にIL-18を分泌することを明らかにした。このように、IL-18は、多様な臓器で多彩な機能を果たしていることを語り始めている。
