培養肝臓星細胞(伊東細胞)による細胞外マトリクス再構築:肝線維化治療の可能性

研究概要	肝臓星細胞は、培養基質の違いにより著しい形態変化を示し、特に、間質型コラーゲンゲルを用いることによりin vivo類似の多数の細胞突起を伸長するように変化する。本研究の結果は、この細胞突起誘導が、細胞表面インテグリン受容体と間質型コラーゲンとの結合、および様々な細胞内シグナル伝達系を介し、最終的に細胞骨格、特に、微小管の再構築によって起こることを明らかにした。また、妹尾らが報告しているように、培養基質の相違により、星細胞の形態変化のみならず増殖能やコラーゲン合成・分泌活性も変化することが明らかにされている。さらに、本研究におけるゼラチンザイモグラフィーによるマトリックスメタロプロテアーゼ(MMP)解析結果は、I型コラーゲンゲルを用いた培養系における活性型MMP-2の存在を示し、これはポリスチレン培養皿内での培養系および基底膜成分を含むマトリゲル上での培養系では認められなかった。しかし、RT-PCRにより、MMP-2に対する阻害剤であるTIMP-2の発現もみられたことから、MMP-2活性はプロセシング後の段階で抑制されていると思われる。一方、in situ zymographyおよび免疫蛍光染色により、I型コラーゲンゲル上での培養系におけるMMP-1の発現が観察された。従って、IV型コラーゲンを含む基底膜成分やI型コラーゲンを含む間質成分など各種細胞外マトリックス成分の再構築が、用いる培養基質の種類に依存して、転写レベルおよびタンパク合成・分泌後の段階で調節されているものと思われる。以上のことから、培養基質として各種細胞外マトリックス成分を用いた肝臓星細胞培養系は、肝組織における細胞外マトリックス再構築のメカニズムの研究に極めて有用であり、コラーゲンなど細胞外マトリックス成分の合成・分泌活性およびMMP発現制御による肝線維化治療の可能性が示唆された。
