TGF-β活性化の制御を介した肝線維化抑制に関する研究

研究概要	肝における細胞外マトリックス産主の中心的役割を担う星細胞(伊東細胞)はtransforming growth factor (TGF)-βにより活性化され、線維合成を亢進させる。このTGF-βは潜在型で分泌される為、一部のペプチドがproteaseの働きにより切り出されて活性型となる。肝硬変では肝においてTGF-βが過剰産生され星細胞を刺激していることが知られており、TGF-β活性化の制御がその治療上1つの戦略目標となる。我々は、星細胞におけるTGF-β活性化のメカニズムの解明を進め、昨年度、既に報告したplasmin以外にもkallikreinなどのproteaseが関与していることを明らかにした。現在更にその同定を試みている。また、これらのproteaseを広域に阻害するprotease inhibitorを20数種の化合物よりスクリーニングし、経口投与が可能で安全性が確立されている2化合物を得報告した。今年度は、臨床導入を目指して慢性肝障害動物モデルでの肝線維化抑制実験を行った。スクリーニングした2化合物の内、既に臨床で使用されているFOY 305をブタ血清誘発肝線維化ラットモデルに投与すると、有意な肝線維化の抑制効果がみられた。今後は、さらに他の肝硬変動物モデルでも有効性を確認する予定である。
