Reg蛋白を介したガストリンによる胃上皮細胞、胃癌細胞の増殖促進機構の解明

研究概要

ガストリンが胃粘膜ECL細胞のCCK-B受容体を介してECL細胞からのReg蛋白の発現を促進しているかを明らかとするためにラットを用いた研究をおこなった。ラットにオスモティックミニポンプを用いてガストリンの持続慢性投与をおこなうと胃のECL裁縫からのRegmRNA、Reg蛋白の産生が促進され、この効果は、CCK-B受容体拮抗薬の同時投与で消失した。
したがって、ガストリンはECL細胞に直接作用して増殖因子であるReg蛋白の産生を促進することが明らかとなった。また、この時に胃粘膜組織の形態学的な検討をおこなうと、Reg蛋白の産生が促進されている場合にのみ胃粘膜の増殖細胞帯での増殖細胞が増加し、胃底腺組織の過形成がみとめられた。
さらに、ヒト生検胃粘膜を用いた検討でも、高ガストリン血症を有する患者ではECL細胞でのRegの産生が亢進しており、血清ガストリン値とReg蛋白産生の間に正の相関があることが明らかとなった。また、高ガストリン血症と高Reg発現を示す例の胃体部粘膜は過形成を示していた。
