Reg蛋白を介したガストリンによる胃上皮細胞、胃癌細胞の増殖促進機構の解明

研究概要

前年度の研究よりラットにおいてもヒトにおいてもガストリンが胃粘膜のECL細胞に作用してReg蛋白の産生を促進していることが明らかとなった。そこで本年度は、産生されたReg蛋白が胃粘膜細胞の増殖を促進させうるか否かを明らかにする目的で研究を行った。まず、ラットの胃粘膜を酵素処理することによって単離細胞とし、この中から増殖帯に存在する増殖細胞を用いた胃粘膜増殖帯細胞の培養系を確立した。この培養系にリコンビナントReg蛋白を添加すると、細胞増殖が著明に促進されReg蛋白が胃粘膜細胞の増殖を促進することが明らかとなった。
また、ECL細胞の培養系にReg蛋白の中和抗体を添加するとECL細胞の増殖は抑制され、過剰量のReg蛋白によって増殖は促進された。
これらの成績から、Reg蛋白はガストリンによって胃粘膜のECL細胞から産生され、胃粘膜の増殖をパラクリン オートクリン機構によって促進していることが明らかとなった。
