アルファ-フェトプロテイン遺伝子サイレンサー因子の同定とその活性調節機構の解析

研究概要

α-Fetoprotein(AFP)遺伝子サイレンサー活性は生後のAFP発現の急速な低下に関与するとともに、肝発癌過程ではその活性低下がAFP遺伝子の再発現をもたらすと考えられている。これまでの研究成績から、このサイレンサー活性はAFPエンハンサー領域と関連し、その活性を強力に抑制すること、さらにAFPエンハンサー領域活性の抑制は、AFPのみならずAFPと並立して存在するアルブミン遺伝子の発現に対しても抑制的に作用することが明らかとなった。このサイレンサー機能の誘導において、Retinoblastoma(Rb)geneあるいはRas geneをHuH-7肝癌細胞を用い強制発現させたところ、Rb geneの導入は肝特異的転写因子であるHFN-1の発現増強を介し、AFP及びアルブミンプロモーターの活性を上昇させ、両遺伝子の発現は転写レベルで増加した。一方、Ras geneの導入はAFPエンハンサー活性を直接抑制した。いずれの遺伝子導入も直接的にはサイレンサー活性を誘導しなかったが、Rb遺伝子の肝癌細胞への導入は、細胞のコンタクトインヒビッションの誘導とともにHNF-1を介する肝特異的遺伝子の転写を増強させ、肝癌細胞の分化を誘導し得るものと考えられた。また、遺伝子のループ形成に関連するHMG遺伝子群の肝癌細胞における発現レベルをRT-PCRを用い検討したところ、HMGI-C遺伝子の発現はAFP高産生肝癌細胞株でAFP低産生株に比較して明らかに高く、HMGI-Cがサイレンサー機能と関連する可能性が示唆された。さらに、これらの研究成果を踏まえ、AFP5'上流転写調節領域の制御下に自殺遺伝子に発現するベクターを作成し、培養癌細胞に導入したところ、in vitro、in vivoにおいて肝癌特異的に抗腫瘍効果が得られた。これらの結果は今後肝癌の遺伝子治療を組み立てる上で重要な視点を与えるものと思われる。
