喘息治療薬における気管平滑筋細胞Ca^<2+>依存性K^+チャネルの意義

研究概要	気管支喘息治療薬の気管平滑筋細胞Ca^<2+>依存性K^+チャネルに対する意義をさらに検討した。既に、喘息治療薬として広く用いられているβ受容体刺激薬の弛緩作用には、モルモット気管平滑筋のみならずヒト気管平滑筋においても、Ca^<2+>依存性K^+チャネルが気管平滑筋の収縮、弛緩に係わることを明らかにすることができた。我々はβ受容体刺激薬の気管平滑筋弛緩作用には、Ca^<2+>依存性K^+チャネルの活性化が伴っていることをすでに明らかにしているが、加えてβ受容体の反応性の低下にはCa^<2+>依存性K^+チャネルの低下が関与し、cAMP依存性のリン酸化を介する効果でないことを示唆する成績が得られた。さらに、adenylate cyclaseを直接活性化することが知られている水溶性forskolinであるNKH477とguanylate cyclaseを介して作用するuroguanylinが気管支喘息患者の治療薬としての可能性を示唆する結果を得ることができた。NKH477についても、モルモット気管平滑筋を用いて、Ca^<2+>依存性K^+チャネルがその効果に係わる結果も得ている。これらの知見より、喘息治療薬におけるCa^<2+>依存性K^+チャネルの意義の一部を明確にできたものと考えている。これらの成績は、Am.J.Respir.Crit.Care Med.,Br.J.Pharmacol.,Eur.J.Pharmacol.およびLife Sci.などの欧米の一流誌に報告した。我々は、これらの成果を踏まえて、新しい気管支喘息治療薬の開発を目指した研究をさらに発展させる。
