肺損傷後のリモデリングにおけるシクロオキシゲナーゼ発現の分子生物学的解析

研究概要	肺の線維化は肺組織の非可逆的変化であり、組織傷害に引き続く治癒過程とも考えられ、進行性の場合は予後不良である。線維芽細胞増殖は繊維化の過程において重要な変化であり、増殖調節の解明は治療法確立のため重要である。PGE2は線維芽細胞増殖を抑制する代表的メディエーターであることより、繊維化発症の機序解明を目的とし、線維芽細胞、単球/肺胞マクロファージ(AM)からのPGE2産生をTh1,Th2様細胞産生のサイトカインによる調節の面より検討した。LPS刺激単球/AMからのPGE2産生は、COX2の発現量に依存的に増加し、その産生量はAMが単球に比し高値であった。Th2様サイトカイン(IL-4,10,13)をこの系に加えるとPGE2産生は抑制され、その程度はIL-4が最も強かった。単球に比べAMでは抑制の程度は軽度であった。一方Th1様サイトカインであるIFNγを加えてもPGE2産生に影響を与えなかった。またTh2様サイトカインはPGE産生と同様にCOX2の発現をmRNA、蛋白レベルで抑制した。これらの結果よりTh1様,Th2様細胞由来のサイトカインは単球/AMからのPGE2産生調節を介して線維芽細胞増殖に関与し線維化病態形成に重要な役割を演じている可能性が示唆された。次に病変部の線維芽細胞のPGE2産生を含む機能変化を検討するため、ラットにブレオマイシン肺臓炎を作成し線維芽細胞採取を行い細胞株を樹立した。肺臓炎モデルから採取した線維芽細胞は対象に比べ強い増殖能を示した。
