パラインフルエンザウイルス4型の全遺伝子解明とその分子疫学的研究

研究概要

ヒト型パラインフルエンザウイルス4型の遺伝子解析および分子疫学的、血清疫学的解析を行った。まず現在までに解析が行われていなかった4B型ウイルスのHN蛋白質の遺伝子解析を完了した。またこの解析結果から4A型ウイルスのHN蛋白質と糖鎖結部位の配列が2箇所異なることが判明した。point mutation法により4B型ウイルスのHN蛋白質cDNAを4A型ウイルスのHN蛋白質に変えたものを3つ作製し、これらをL929細胞に構成的に発現させ、flow cytometry法でモノクローナル抗体を用いて、抗原性を解析したところ、2箇所とも4AウイルスのHN蛋白質と同じにしたクローンの抗原性は4A型ウイルスのHN蛋白質と似た抗原性を示したが、完全には一致しなかった。4型ウイルスのみサブタイプに分かれるが、HN蛋白質の糖鎖が関与している可能性が示された。残されたL蛋白質の遺伝子は一部解析を行ったが完全解明には至らなかった。また小児の血清につき、モノクローナル抗体を用いて、4型ウイルスのを検出したところ数パーセントが陽性であったが、2型ウイルスなどとの交差反応も考えられ、さらなる解析が必要と思われた。
近年ウイルスレセプターや細胞融合に関与する細胞側の因子としてCD98/4F2/FRP-1分子やインテグリンシステムが注目されている。本研究でもこれらに注目し、まずCD98分子の4型ウイルス増殖との関連を解明すべく、動物モデル作製のため、ヒトCD98HcDNAをベクターpS65T-C1に組み込み、日本SLCとの共同研究でtransgenicマウス作製にとりかかった。現在マウスの分娩が終わり90匹の子供が誕生し、近々PCR法で解析の予定である。これらのマウスにつき感染実験にて、抗体、ウイルス遺伝子の有無を調べる予定である。
