Tripret repeat disease遺伝子(特にMJD1gene)の不安定性のメカニズムについて

研究概要

我々はこれまで、Machado-Joseph病(MJD)の原因遺伝子(MJD1)内のCAGリピートの不安定性について、1)病気が父親由来では、母親由来であるときに較べて大であること、2)正常アリル(NA)のCGG/GGG多型が、増大アリル(EA)のCAGリピートの不安定性に大きく影響を与えること(inter-allelic interaction)、3)病気が父親由来の時には、子供の世代での発症者の比率が高いこと(meiotic drive)をspermのレベルで照明して報告してきた。今年度、我々はトリプレット・リピート病のなかでも最も著明な表現促進現象を示すDRPLAについて、原因遺伝子内のCAGリピートが不安定である機序を解明するために、DRPLA患者2名の計316個のsingle spermにおけるCAGリピート数の解析を行なった。その結果、1)増大アリル(EA)と正常アリル(NA)の分離比は1:1からの有意な偏りは認めず、meiotic driveの現象を支持することはできなかった。2)NAは非常に安定であった。3)リピートの不安定性を現す最も良い指標とされているEAの分散は51〜316であり、他のCAGリピート病に比し大きかった。4)患者1のEAの分散は患者2のそれに比し、有意に大きく(F-test、P<0.0001)、更にこれまでに報告さているCAGリピート病のなかでも際だって大きかった。DRPLAは小脳失調の他に、痴呆や精神症状を呈するので、spermの入手はきわめて困難であり、今回の我々のデータは大変貴重である。現在、患者1と患者2のリピートの不安定性の違いに着目して、不安定性に関与するcisやtransのfactorが存在するのか否かの検討を行なっている。今後、CAGリピート病に共通する不安定性の機序につき検討する予定である。
