皮膚における内因性抗炎症物質の同定とその発現調節機序の解析

研究概要	1 ステロイド長期外用による接触皮膚炎の増強反応の誘導昨年に引続き、Balb/Cマウス背部皮膚に隔日にステロイド懸濁液を9回塗布する長期塗布モデルマウスの皮膚反応の亢進機序を検討した。昨年CD8陽性細胞がこの増殖反応に関与することを報告したが、この系において所属リンパ節にγδ陽性細胞が著増していることを見出した。この細胞の誘導が皮膚反応の増強に関与していると考えられた。また外用ステロイドの長期塗布は、IgE依存性も皮膚反応も増強したことより皮膚における炎症をその誘因に拘わらず増強すると考えられた。 2 ステロイド長期外用マウス皮膚におけるサイトカインの動態上記ステロイド長期連用マウスに対し、凍結融解にて得たケラチノサイト由来蛋白と局所投与し、その効果を検討したところ、惹起反応の抑制が見られたことより、ステロイド長期連用によりケラチノサイト由来炎症抑制物質の産生低下あるいは炎症増幅物質の産生増強の可能性が考えられた。このため、IL1ra,IL10を投与したところ遅発相の皮膚反応が抑制された。炎症局所においてもmRNA、蛋白レベルでIL10の発現低下、IL1αの発現増強が見られたことよりケラチノサイト由来のIL10の産生が内因性抗炎症物質として重要であると考えられた。 3 培養皮膚ケラチノサイト由来物質による増幅皮膚反応の制御 2の効果を検討するために、ハイドロコーチゾンとハプテン(DNBS、TNBS)を添加した培養系でケラチノサイトを培養し、その培養上清中のサイトカインを測定した所、IL1α、IL6の著明な産生増強を認め、NFκBの発現増強を認めたことより、炎症反応の増強にNFκBの関与が考えられ、逆にIL10がその制御に関与していると考えられた。今後IL10のNFκBの発現への影響の検討が重要と考えられた。
